  • Developed a capillary electrophoresis method for Process Analytical Technology (PAT), forced degradation, and developmental stability studies conducted according to ICH guidelines
  • Developed a stability indicating LC-MSn methods for key degradation pathways such as oxidation and deamidation of plasma derived proteins
  • Developed LC-MS-based methods for recombinant protein therapeutics. Studies included post-translational/process related modifications, mapping of disulfide bonds, and peptide sequence mapping

Research Assistant, 2001-2006, Department of Chemistry, Purdue University, IN

The research was directed towards characterizing biological molecules in the gas-phase. The studies involved development of novel instrumentation, ion chemistries, theories, and proteomics applications. The projects are outlined below:

  • Synthesis of protein complexes via gas-phase ion chemistry and mass spectrometry as a novel means of understanding gas-phase protein complexes. Upto six heteomeric protein complexes have been assembled by sequential gas-phase ion/ion reactions
  • Identification of Yeast whole cell proteins and their post-translational modifications by examining low mass-to-charge fragmentation spectra via multi-dimensional chromatography-mass spectrometry and data base search tools
  • Investigation of the charge state dependennce on the cleavage of disulfide-linkages of whole protein ions due to ion/ion electron transfer (e.g. insulin was used as a model protein)
  • Developed a theoretical frame work to predict fragmentation of high-mass bio ions during proton transfer ion/ion reactions. Rice-Ramsperger-Marcus-Kassel Theory (RRKM) and Monete Carlo MD simulations were used to predict unimolecular dissociation and collision cooling rates respectively inorder to model energy deposition in large proteins and their complexes
  • Examined the role of protein pI and solution pH on enhancing the selective ESI response of protein mixtures
  • A predictive model was developed to identify reagent anions for their ability to accept a proton or donate an electron to peptide cations during Electron Transfer Dissociation (ETD). The likelihood of electron transfer between the reagent anion and the polypeptide cation charge bearing sites were modeled based on the Landau-Zener theory for electron transfer and curve crossing phenomena
  • Examined the utility of sequential ion/ion reactions and MSn capabilities (i.e. MS3: PTR charge inversion followed by ETD to study post-translational modifications such as phosphorylation; MS4: CID of whole protein ions and PTR with ion parking followed by ETD)
  • Designed experiments and molecular dynamics simulations to examine the mechanisms for electron capture dissociation (ECD) and electron transfer dissociation (ETD) of disulfide-linked poly protonated and fixed charge peptides. The findings suggests that through-bond electron transfer plays a role besides the accepted Hot-hydrogen atom mechanism
  • Developed a novel gas-phase reaction for selective cleavage of disulfide-linked polypeptides via transition metal-peptide complex. This is the first selective ion/ion dissociation reaction reported using AuCln anions

Research Assistant, 1999-2001, Department of Chemistry, University of North Carolina, NC

The research was directed towards understanding solution phase binding interactions of protein small molecule ligands. The projects are outlined below:

  • Mass Spectrometry and Affinity Capillary Electrophoresis (ACE) were used to delineate the nature of protein-sodium dodecyl sulfate (SDS) binding
  • Developed a CE-MS sheath flow interface to correlate gas-phase protein-SDS binding with solution phase protein-SDS binding
  • Developed LC-MS methods for small molecules for cooperate clients at the Regional Analytical Chemistry Laboratories
