C1 Side

New technology now allows for efficient and high-throughput gene expression analysis from single cells.   C1 Single-Cell Autoprep System is now offered through the AA Core and allows users to rapidly and reliably isolate, process, and profile up to 96 individual cells for genomic analysis (qPCR or RNAseq), allowing you to study cell differentiation, measure individual cell responses to specific stimuli, verify critical disease biomarkers, validate RNAi knockdown, and conduct candidate drug screens. You can extract, reverse transcribe, amplify, and ultimately detect and analyze gene expression using just one technology, eliminating the added variability introduced by human error and multi-platform approaches. With an optimized protocol, pre-formulated reagent kits, and disposable IFCs, users can achieve single-cell accuracy with “load and go” productivity and a cDNA product that is ready for use with the AA Core’s BioMark HD high throughput PCR system.

Single-Cell Isolation and Specific Target Amplified (STA) cDNA Preparation: Single cell suspensions from cell culture, primary isolates, or FACS may be submitted to the AA Core for processing on the C1 Single-Cell Auto Prep IFC. Each of these chips is optimized for isolating cells of a certain size, so it is critical that users have an accurate assessment of their average cell size or obtain one from the Core to ensure proper recovery.

Single-Cell Isolation and Whole Transcriptome cDNA Preparation: Single cell suspensions from cell culture, primary isolates, or FACS may be submitted to the AA Core for processing on the C1 Single-Cell Auto Prep RNAseq IFC. Each of these chips is optimized for isolating cells of a certain size, so it is critical that users have an accurate assessment of their average cell size or obtain one from the Core to ensure proper recovery. The C1 will automatically generate and amplify cDNA to sufficient levels for Illumina library preparation.  The AA core will submit your cDNA samples to the UNC Genomics Core where Illumina libraries will be prepared and sequenced on a HiSeq2000.

Submitting Samples:

To begin a project using the AA Core’s C1 system, pleasC1 Traye contact the Core Manager to arrange a consultation. Once project details have been coordinated, samples may be submitted. All samples should be dropped off in Room 030 of the Glaxo Building, and new users are advised to make an appointment to ensure that someone will be available to receive their samples.
