Cyan Troubleshooting Guide

  1. A tube is running but no events are appearing on the screen.
    1. Make sure that autosample is checked and that a tube is being detected.
    2. Make sure that you are using a polypropylene tube.
    3. Check for a clog. Do a sample clean with water and make sure the level of water is decreasing. If it is not decreasing, the instrument is clogged. Call the flow core.
    4. Check for a software error. Try creating a new database and protocol.
    5. Refresh the count after boost with the clear tab or 'control z'
    6. Place blue QC beads on the instrument and run. You should see events about the size of lymphocytes and green fluorescence. 
  2. There is a pressure or vacuum error.
    1. Hit 'Reset' on the fluidics cart. 
    2. Click “Startup” again to put the instrument through the startup process.
  3. An error message with “DCM” pops up.
    1. Restart the computer.
  4. The data does not look “normal”.
    1. Verify that your samples were filtered before running (not just before staining). For cell filtration options, see: https://www.med.unc.edu/flowcytometry/resources/application-protocols/cell-filtration-options-1
    2. Check for a clog. Do a sample clean with water and make sure the level of water is decreasing. If it is not decreasing, the instrument is clogged. Call the flow core.
    3. Check for a software error. Try creating a new database and protocol.
    4. Trigger on Side Scatter instead of Forward Scatter
  5. The microtiter tube within a tube is not working.
    1. Try a new outer tube. Sometimes these can get worn down and will not seal properly.
    2. Make sure that the probe is not hitting the bottom of the microtiter tube.
    3. Increase the pressure bar. (the pressure bar on Thurston and LCCC will have to be higher than MEJ to run the microtiter tubes) If the pressure bar is not high enough, the tube will not run. 
  6. To Toggle Backflush off and on
    1. In Summit go to the Edit command. 
    2. Then under Instrument select Cyan. 
    3. From there a dialog box will show preferences that can be edited. 
    4. One of the boxes to select is "Auto Backflush". 
    5. When selected the Cyan will automatically backflush when a tube is not detected. 
    6. When unselected you will have to manually backflush from the normal instrument control panel.
