Ph.D. Project Description:
The Role of Vascular Endothelial Growth Factor (VEGF) signaling in the Regulation of Small GTPase Rac1 Exchange Factors
Principle Investigator:
Dr. Keith Burridge

IVB Collaborator:
Dr. Susan Smyth
Project Description:
We plan to use endothelial cells from mice lacking the expression of specific Rac1 exchange factors to determine if they are required for blood vessel formation.

Rotations:
Steve Clarke: The goal of this project was to understand how a protein called the Smith anigen (Sm antigen) initiates abberant B-cell production during an auto-immune response in diseases such as lupus.
Vicki Bautch: The aim of this project was to elucidate how vascular endothelial growth Factor (VEGF) and Sonic Hedgehog (Shh) mediate endothelial cell differentiation and tissue formation during embryogenesis by measuring tissue-specific gene expression with RT-PCR.
Keith Burridge: In this project, I studied the role of small GTPase RhoA and its downstream effector Rho Kinase (ROCK) on leukocyte transmigration across endothelial lining of blood vessel walls.

Familiar Techniques:
Western Blotting PCR Immunoflourescence Subclobing and gene expression
Other Techniques used in Laboratory:
Small GTPase Activity Assays Guanine Nucleotide Exchange Factor (GEF) Pulldown Assays Chemotaxis and Migration Assays
