<p>The Cancer Biology Group at NIEHS focuses on early events in skin tumor development using a transgenic mouse model (TgAC). This model possesses a v-Ha-ras transgene under the regulation of a fetal globin promotor integrated at an ectopic site which confers a unique phenotype of inducible skin papillomas with a high rate of progression to invasive squamous and spindle cell neoplasms. The goals of our studies are to identify and characterize: 1) The cellular origin of the tumors and 2) critical genes which are involved in ras-mediated tumor induction and progression. Conventional cancer therapies have until recently depended on treatment of late stages of tumor growth and involved non-specific mechanisms of cellular injury. By focusing on understanding early events in tumor induction we hope to gain insights into targets for intervention that can more specifically inhibit cancer cell growth.</p>]]></content:encoded>

<p>The research in our laboratory involves several major projects related  to the molecular pathogenesis of human cancer and investigations related  to the biology of liver stem-like progenitor cells, including (i)  characterization of human liver tumor suppressor genes, (ii) analysis of  genetic determinants of breast cancer, (iii) investigation of  mechanisms governing aberrant DNA methylation in breast cancer, (iv)  liver progenitor cell responses after toxic liver injury, and (v)  transplantation of liver stem-like progenitor cells for correction of  genetic liver disease.</p>]]></content:encoded>

<p>1) Using cellular stress signaling pathways to develop in vitro assays  in human cells to rapidly ascertain the toxic potential of a wide range  of environmental compounds including nanoparticles used in commerce.  This research draws extensively on signaling pathways in cells and on  high throughput methods including robotics and developing methods for  data analysis. 2) Design molecular studies to ascertain the mode of  action of environmental compounds of potential concern. This research  employs the use of gene over expression and suppression (RNAi) in  established and primary cell cultures. 3) Development of alternate  species assays for in vivo rapid screening of environmental compounds  using normal and transgenic zebrafish.</p>]]></content:encoded>

<p>Research in my laboratory is directed toward achieving a better understanding of the mechanisms and pathogenesis associated with a variety of environmentally induced or genetically based birth defects. This information is then applied to development of preventative/ameliorative measures relative to these defects. Our interest in modeling human genetic malformation syndromes and opportunities for collaborative efforts with molecular geneticists who have produced transgenic mice and mice with targeted gene modification have proven productive in our attempt to better understand the developmental basis for a variety of malformations of the brain including anencephaly, holoprosencephaly, and hydrocephaly. Regarding teratogen-induced birth defects, our major emphasis is on Fetal Alcohol Spectrum Disorders (FASD). Currently, high resolution magnetic resonance imaging (MRI) is being utilized to identify, characterize, and correlate the craniofacial, ocular, otic and CNS dysmorphology that results from prenatal ethanol exposure at specific stages of embryogenesis. These studies are designed to inform human clinical research and to expand the diagnostic criteria for prenatal alcohol exposure.</p>]]></content:encoded>
