Nucleotides: All natural base phosphoramidites are commercially available. We do synthesize deoxy oligos, and others with a variety of 2’-modifications such as 2’-OMe, 2’-O, 4’-C-locked nucleic acid (LNA), 2’-F etc. We are also able to synthesize oligos with fluorescent dyes (such as TAMRA, 6-FAM, Yakima Yellow, Teas Red X etc) at either 3’ or 5’ terminus according to your need.
Sequence and length: Oligonucleotide synthesis is started on the CPG (controlled pore glass) support from 3’ terminus with the removal of DMTr group. We believe we can provide you with high quality oligos up to 60 mers, however it is possible to make oligos with greater length (> 60 mers), but purification could be a challenging task.
Backbone Modification: Backbone modification is crucial for certain biological properties of oligos. We can provide you oligo with either phosphodiester ( PO) or phosphorothioate (PS) backbones upon your request (as well as PNAs with the Expedite).
Linker: Linkers could be useful for postsynthetic modification of oligos to conjugate peptides or other substances. There are some linker phosphoramidites commercially available. The linkers include carboxy modifier C10, PC amino modifier phosphoramidite, amino modifier C6, C7, C12, thiol modifier C6 S-S, thiol modifier C6 etc. For details see linker information in the Glen Research catalogue. We can incorporate linkers at either 3’- or 5’-end of the oligo according to your request.
Quantity: We perform synthesis on our latest version of ABI 3400 instrument between 40 nanomole to 1 micromole scale. We can synthesize up to 4 micromoles at a time.. We can scale up the synthesis upon request and can double or triple the synthesis to fulfill your need.
Cost: The cost for oligos varies upon synthesis scale, type of modification and length of oligo you are ordering. Please see the pricing table. This includes lyophilization, the HPLC purification (>99%) and MALDI-TOF mass analysis of the purified oligo. Cost for some special oligo synthesis will be higher than usual depending upon incorporation of fluorescent dye, DNP labeling, amino modifier, thiol modifier or other alterations. Our current policy is that the Core will cover costs of small batches of oligos from its supply budget as far as possible. However, larger batches (>1 uM) will need to be paid for from the individual PPG projects.
Time: Synthesis of oligonucleotide is FAST, it takes about only couple to several hours depending upon the length and extra coupling time for modification. However, cleavage, deprotection, lyophilization, HPLC purification followed by further lyophilization, and analysis by MALDI-TOF mass spectroscopy requires a considerable time to have a high quality oligo. Although these are routine for oligo making, we are able to deliver oligo within two to three weeks. Delivery might be more delayed if you require many milligrams oligo.
