Seminar title: “Establishing the relationship between chromatin accessibility and preparation for DNA replication”

Abstract: The process of preparing chromosomes for replication takes place during the G1 phase of the cell cycle by loading chromosomes with precursors to the replicative DNA helicase. This process is a prerequisite for normal DNA replication in the following S phase. However, it is not yet understood how cells know they are fully prepared for S phase entry or how they organize this process to avoid large unreplicated regions. In this study we combine quantitative live cell and fixed cell imaging to analyze helicase precursor loading relative to chromatin markers.

Seminar Title:“Structural and dynamic basis for light-induced allosteric control of an engineered Rho family GTPase”

ABSTRACT

GTPases play a vital role in many cellular signaling processes and are recently engineered to respond allosterically to light via insertion of a LOV domain. To better understand the allosteric mechanism, we are using structural and biochemical approaches to characterize how these proteins respond to light stimulation. Specifically, NMR spectroscopy is used for detecting differences in structure and dynamics for “dark” and “lit” states.
