“Selective ER-Export of Misfolded Secretory Pathway Proteins for Lysosomal Degradation”

ABSTRACT
We discovered that a subset of misfolded secretory pathway proteins, including variants of prion protein and amyloid precursor protein, are selectively exported out of the ER by a constitutive and stress-enhanced pathway named RESET. After RESET, these misfolded proteins are subsequently degraded in lysosomes. We now present our new unpublished findings on the mechanism by which misfolded proteins are constitutively released for degradation through RESET and how ER stress enhances this pathway. Implications for protein misfolding diseases will be discussed.
