Due to their essential roles in gene regulation, miRNAs are tightly regulated, and abnormal miRNA expressions have been linked to cancer, neurological disorders, cardiovascular diseases, and other diseases.<\/p>\n<p>Understanding how miRNAs are regulated has been the focus of intense recent study. Over the years, scientists have discovered multiple protein regulators of miRNA biogenesis cellular pathways, and it is often perceived that these protein factors act on largely passive miRNA processing intermediates to direct their maturation. Now, the lab of\u00a0<a href=\"https:\/\/www.med.unc.edu\/biochem\/directory\/zhang\/\">Qi Zhang, PhD<\/a>, associate professor in the UNC Department of Biochemistry and Biophysics, discovered a new RNA-centric mechanism by which miRNA processing intermediates can play direct, active roles in regulating miRNA biogenesis.<\/p>\n<figure id=\"attachment_4307\" class=\"thumbnail wp-caption alignright\" style=\"width: 210px\"><a href=\"https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2018\/07\/jared-baisden.jpeg\"><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-4307 size-full\" src=\"https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2018\/07\/jared-baisden.jpeg\" alt=\"Jared Baisden\" width=\"200\" height=\"200\" srcset=\"https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2018\/07\/jared-baisden.jpeg 200w, https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2018\/07\/jared-baisden-150x150.jpeg 150w, https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2018\/07\/jared-baisden-64x64.jpeg 64w\" sizes=\"auto, (max-width: 200px) 100vw, 200px\" \/><\/a><figcaption class=\"caption wp-caption-text\">First author Jared Baisden, PhD, a recent alumni of Qi Zhang lab and BCBP<\/figcaption><\/figure>\n<p>These findings,\u00a0<a href=\"https:\/\/www.nature.com\/articles\/s41589-020-00667-5\">published in the journal\u00a0<em>Nature Chemical Biology<\/em><\/a>, unveil that a \u2018hidden\u2019 layer of regulation by which the intrinsic dynamic ensemble of miRNA processing intermediates can direct the outcome of important biological processes in response to environmental and cellular stimuli in the absence of protein factors. If these processes go awry, then disease could result. Understanding the roles of miRNAs in disease is a needed step in finding new routes to better therapeutics.<\/p>\n<p>MicorRNA-21 (miR-21) is involved in the creation, progression, metastasis of cancerous tumors, and it is involved in cell survival. When Zhang\u2019s lab studied miR-21 using NMR relaxation dispersion \u2013 an advanced imaging technique capable of detecting sparsely populated transient states that often evade conventional detection techniques \u2013 they found the precursor of miR-21 exists as an ensemble of dynamic conformational states, which is surprisingly sensitive to the acidity of the environment.<\/p>\n<p>They discovered that, across physiological conditions, about 1 \u2013 15% of the miR-21 precursor carries one additional proton, the smallest chemical modification known as protonation. With a lifetime of approximately 0.8 milliseconds, this protonated state sequesters a key residue into a new structure that substantially enhances the efficiency of processing the precursor into mature miR-21.<\/p>\n<p>With these powerful \u201cimaging\u201d techniques, scientists can now unveil transient RNA states as the \u2018hidden\u2019 layers of regulation, including a fleeting riboswitch state important in controlling gene transcription \u2013 a state the Zhang laboratory discovered in 2017.<\/p>\n<p>\u201cWe think these techniques add more promise for new strategies to create RNA-targeted therapeutics,\u201d said Zhang, a member of the UNC Lineberger Comprehensive Cancer Center. \u201cAnd that is our goal; we need better targeted therapies for many diseases including cancers.\u201d<\/p>\n<p><em>Other authors of the Nature Chemical Biology paper are Jared Baisden, PhD, a recent graduate in the Zhang lab; Joshua Boyer, PhD, a postdoctoral fellow in the Zhang lab; Bo Zhao, PhD, a former graduate in the Zhang lab; and Scott Hammond, PhD, associate professor in the UNC Department of Cell Biology and Physiology.<\/em><\/p>\n<p><em>This work was funded by the National Science Foundation and a Jefferson-Pilot Award from the UNC School of Medicine.<\/em><\/p>\n<hr class=\" rule-dotted osc-rule\" \/>\n<p>News courtesy of UNC Health.
