Proteins that control gene expression and repair of DNA damage must bind to\u00a0<\/span><span data-contrast=\"none\">nucleosomes<\/span><span data-contrast=\"none\">\u00a0to perform their functions. While studies of individual proteins bound to the\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0have shown that an acidic patch made of negatively charged atoms on the\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0may be a common site for protein binding, the pervasiveness of this acidic patch binding and whether\u00a0<\/span><span data-contrast=\"none\">other<\/span><span data-contrast=\"none\">\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0binding\u00a0<\/span><span data-contrast=\"none\">hot-spots<\/span><span data-contrast=\"none\">\u00a0exist remained unclear. This study describes an elegant proteomics screen that establishes the universal principles of\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0recognitio<\/span><span data-contrast=\"none\">n.\u00a0<\/span><span data-contrast=\"none\">The authors prepared a library of nucleosomes,<\/span><span data-contrast=\"none\">\u00a0each containing a cluster of mutations within a different patch on the\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0disk surface. Quantitative mass spectrometry was then used to compare the abundance of proteins that interact with each of the\u00a0<\/span><span data-contrast=\"none\">nucleosomes<\/span><span data-contrast=\"none\">\u00a0in the library. By analyzing how each cluster of mutations influenced\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0binding proteome-wide,\u00a0<\/span><span data-contrast=\"none\">the authors paired hundreds<\/span><span data-contrast=\"none\">\u00a0o<\/span><span data-contrast=\"none\">f\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0binding proteins with the specific\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0surface requirements.\u00a0<\/span><span data-contrast=\"none\">The\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0acidic patch,<\/span><span data-contrast=\"none\">\u00a0was critical for\u00a0<\/span><span data-contrast=\"none\">the majority of<\/span><span data-contrast=\"none\">\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0binding proteins, often in concert with two adjacent, newly identified hot-spots. This comprehensive\u00a0<\/span><span data-contrast=\"none\">nucleosome<\/span><span data-contrast=\"none\">\u00a0<\/span><span data-contrast=\"none\">interactome<\/span><span data-contrast=\"none\">\u00a0screen uncovers the general rules governing recognition of the fundamental unit of the genome.<\/span><span data-ccp-props=\"{}\">\u00a0<\/span><\/p>\n<figure
