{"id":9339,"date":"2019-05-02T12:05:15","date_gmt":"2019-05-02T16:05:15","guid":{"rendered":"https:\/\/www.med.unc.edu\/biochem\/?p=9339"},"modified":"2019-11-11T11:05:15","modified_gmt":"2019-11-11T16:05:15","slug":"nih-awards-nearly-2-million-to-cell-microsystems-to-boost-biomedical-research","status":"publish","type":"post","link":"https:\/\/www.med.unc.edu\/biochem\/news\/nih-awards-nearly-2-million-to-cell-microsystems-to-boost-biomedical-research\/","title":{"rendered":"NIH awards nearly $2 million to Cell Microsystems to boost biomedical research"},"content":{"rendered":"<p><a href=\"https:\/\/cellmicrosystems.com\/\">Cell Microsystems<\/a>, Inc., a company based in the Research Triangle Park, has received two federal Small Business Innovation and Research awards totaling $1.9 million to automate distinct CRISPR workflows using their proprietary\u00a0<a href=\"https:\/\/cellmicrosystems.com\/cellraft-technology\/\">CellRaft Technology<\/a>, which was created by the lab of Nancy Allbritton, MD, PhD, Kenan Professor and Chair of the UNC\/NCSU Joint Biomedical Engineering Department.<\/p>\n<p>Nick Trotta, PhD, Director of Genomics Programs, described the awards saying, \u201cThese grants will accelerate our on-going work toward automating CRISPR workflows using the CellRaft AIR\u2122 System that will provide a more efficient alternative to other cloning and sorting technologies.\u201d<\/p>\n<p>Cell Microsystems\u2019 CellRaft AIR\u2122 System is an automated platform employing an integrated fluorescent microscope to image, sort, and isolate cells or small clonal colonies cultured on the CytoSort\u2122 Array.\u00a0 Culturing viable, single cells after applying guide RNAs, then tracking clonal colony formation and other phenotypes for screening, gives the System tremendous value in CRISPR workflows.<\/p>\n<p>Under a Phase II grant, the Company will collaborate with two investigators to validate the AIR\u2122 System in core laboratories:\u00a0<a href=\"https:\/\/www.med.unc.edu\/biochem\/directory\/marzluff\/\">William Marzluff<\/a>, PhD, Kenan Distinguished Professor of Biochemistry and Biophysics with a joint appointment in Biology at the UNC School of Medicine, and Mike McConnell, PhD, of the University of Virginia.<\/p>\n<figure id=\"attachment_9349\" class=\"thumbnail wp-caption alignnone\" style=\"width: 310px\"><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-9349 size-medium\" src=\"https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2019\/04\/William-Marzluff-Passport-cropped-10052015_003-2019-300x269.jpg\" alt=\"photo of William Marzluff, PhD distinguished faculty of biochemistry and biophysics and joint with biology\" width=\"300\" height=\"269\" srcset=\"https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2019\/04\/William-Marzluff-Passport-cropped-10052015_003-2019-300x269.jpg 300w, https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2019\/04\/William-Marzluff-Passport-cropped-10052015_003-2019-768x688.jpg 768w, https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2019\/04\/William-Marzluff-Passport-cropped-10052015_003-2019-1024x917.jpg 1024w, https:\/\/www.med.unc.edu\/biochem\/wp-content\/uploads\/sites\/795\/2019\/04\/William-Marzluff-Passport-cropped-10052015_003-2019.jpg 1200w\" sizes=\"auto, (max-width: 300px) 100vw, 300px\" \/><figcaption class=\"caption wp-caption-text\">William (Bill) Marzluff, PhD, Kenan Distinguished Professor of Biochemistry and Biophysics<\/figcaption><\/figure>\n<p>Marzluff, who is also a member of the UNC Lineberger Comprehensive Cancer Center said, \u201cThe AIR\u2122 System helps us identify a much larger number of viable clones for generating knockout cell lines, and large numbers of cloned cells can be identified and expanded much faster than with other approaches.\u201d<\/p>\n<p>McConnell\u2019s laboratory will use the System to run a pooled CRISPR screen with time-course imaging.\u00a0 \u201cWith the AIR System now in the stem cell core, we can use it for some of the CellRaft applications we\u2019ve already published in single cell and single nucleus sequencing, but we are doing a lot of CRISPR work with it now too.\u00a0 It\u2019s an easy way to track cells for your phenotype of interest, keep them alive, and then collect the cells you want for downstream molecular analysis,\u201d McConnell said.<\/p>\n<p>In a separate Phase I grant, the Company will collaborate with William Buchser, PhD, of Washington University in St. Louis to develop pooled CRISPR-mediated genome editing.\u00a0 \u201cThe AIR System gives us a great way to edit phenotypically interesting cells and track them for subtle functional changes we can only see under optimized culture conditions. We ran a few proof-of-concept studies with the System and it is going to be an excellent platform for functional genomics experiments.\u201d<\/p>\n<p>The Company also announced a new software release for the AIR\u2122 System and a new HexaQuad\u2122 Array. The new software automates cloning workflows using brightfield imaging to eliminate the need for fluorescent markers for sorting and confluency measurement.\u00a0 Another new feature automates cell sorting based on fluorescent signal intensities to characterize complex phenotypes.<\/p>\n<p>James Kane, Director of Sales, said \u201cThe improvement of the fluorescent intensity and brightfield algorithms will empower our customers to monitor single cell culture and isolate clones at the ideal timepoint.\u201d The HexaQuad Array is designed to run 24 distinct experiments simultaneously on a single consumable.\u00a0 Kane added, \u201cThe HexaQuad is uniquely suited to labs who perform high-throughput CRISPR editing in single guide or guide pools, scaling to hundreds of clones per week, and significantly reducing a laborious two-month workflow. This new array coupled with new software provides a five-day workflow at higher fidelity and lower cost than traditional methods.\u201d<\/p>\n<p>In 2010, Allbritton and two members of her lab \u2013 senior scientist Christopher Sims, MD, and research associate Yuli Wang, PhD \u2013 founded Cell Microsystems.
