Simply provide your single-cell suspension and compatible primary antibodies. The AA Core will perform the services and return fluorescence images that can be used to quantify protein expression. The Core will assist with software analysis. Milo is a powerful tool both alone and in combination with the Core\u2019s other single cell services: 10x Genomics, <a href=\"https:\/\/www.med.unc.edu\/cgibd\/cores\/advanced-analytics\/single-cell-autoprep\/\">Fluidigm C1<\/a>, and <a href=\"https:\/\/www.med.unc.edu\/cgibd\/cores\/advanced-analytics\/biomark-hd\/\">Fluidigm BioMark HD<\/a>.<\/p>\n<p>Applications of the Milo system include:<\/p>\n<ul>\n<li>scRNA-Seq validation<\/li>\n<li>Cell subtype, protein isoform, and phospho-protein heterogeneity identification<\/li>\n<li>Histone modification detection<\/li>\n<li>Gene editing efficiency determination<\/li>\n<li>Protein expression in low-abundance samples<\/li>\n<\/ul>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-3185\" src=\"https:\/\/www.med.unc.edu\/cgibd\/wp-content\/uploads\/sites\/466\/2017\/12\/Image-5_Milo-scWest-chip.png\" alt=\"\" width=\"1200\" height=\"469\" srcset=\"https:\/\/www.med.unc.edu\/cgibd\/wp-content\/uploads\/sites\/466\/2017\/12\/Image-5_Milo-scWest-chip.png 1200w, https:\/\/www.med.unc.edu\/cgibd\/wp-content\/uploads\/sites\/466\/2017\/12\/Image-5_Milo-scWest-chip-300x117.png 300w, https:\/\/www.med.unc.edu\/cgibd\/wp-content\/uploads\/sites\/466\/2017\/12\/Image-5_Milo-scWest-chip-768x300.png 768w, https:\/\/www.med.unc.edu\/cgibd\/wp-content\/uploads\/sites\/466\/2017\/12\/Image-5_Milo-scWest-chip-1024x400.png 1024w\" sizes=\"auto, (max-width: 1200px) 100vw, 1200px\" \/><\/p>\n<p>Cell capture is completed by placing 1 mL of suspended cells (10,000-100,000 cells) over a single-cell Western (scWest) chip containing thousands of tiny microwells patterned into pre-cast, photoactive polyacrylamide gel. The Milo system then lyses cells, performs rapid SDS-PAGE separation (~1 min), and immobilizes proteins using UV light. No transfer to membranes is required. Loaded chips are probed with an antibody cocktail. Chips are then ready to be imaged. Contact the <a href=\"mailto:carlton_anderson@med.unc.edu\">Associate Director<\/a> to arrange a consultation.<\/p>\n<p><strong>Cell Capture &amp; Antibody Probing<\/strong>: Please provide a single-cell suspension containing &gt;10,000 cells (10,000-100,000 cells\/mL) and up to two Western-validated primary antibodies. Primary antibodies must be generated in different species for multiplexing. The Core provides all other reagents and supplies. <a href=\"https:\/\/www.proteinsimple.com\/antibody\/milo_antibodies.html\">Click here for a selection of Milo-tested antibodies.<\/a><\/p>\n<p><strong>Imaging &amp; Analysis<\/strong>: The Core will provide scWest imaging service for the user.
