During this process the student will take notes on the Talos Arctica protocol handout from the core website and is expected to ask questions.<\/p>\n<p style=\"font-weight: 400\"><strong>Topics covered during the training session include:<\/strong><\/p>\n<ol>\n<li>Alignment of the TEM.<\/li>\n<li>Using the Autoloader.<\/li>\n<li>Low-dose imaging.<\/li>\n<li>Assessing sample quality and ice thickness.<\/li>\n<li>Acquiring and saving images.<\/li>\n<li>Collecting montages.<\/li>\n<li>Data collection using the serialEM GUI and scripts.<\/li>\n<li>Optional topics will be covered depending on the goals of the student and what imaging modality they are using, ie. single-particle cryoEM, tomography, microED, cryoTEM.<\/li>\n<\/ol>\n<p><strong>Part 2:<\/strong> The student will operate the microscope under the supervision of the core staff.<\/p>\n<p style=\"font-weight: 400\"><strong>The student will do the following:<\/strong><\/p>\n<ol>\n<li>Inventory autoloader, load and unload a grid.<\/li>\n<li>Use the flu screen and TIA to quickly look at the grid at low magnification.<\/li>\n<li>Find Eucentric height manually using the stage wobbler.<\/li>\n<li>Take Search, View, and Record images with SerialEM. Save the images as snapshots and mrc format.<\/li>\n<li>Take a low magnification montage of the grid of a using SerialEM<\/li>\n<li>Use the navigator to move the stage to 3 areas of the grid and take images.<\/li>\n<li>Run scripts, map2grids and dose settings.<\/li>\n<li>Setup data collection using BIS method on 3 grid squares.<\/li>\n<\/ol>\n<p><b><span data-contrast=\"auto\">After Training Module<\/span><\/b><\/p>\n<ol>\n<li>Student reviews their notes taken during a training session on their own.<\/li>\n<li><span data-contrast=\"auto\"> Students scheduled an assessment session with the cryoEM Core Staff. Core Staff watch students operate the microscope. Core staff score the students using a <a href=\"https:\/\/www.med.unc.edu\/cryo-em\/wp-content\/uploads\/sites\/1040\/2025\/11\/UNC-CH-Talos-Arctica-CryoTEM-Certification.pdf\">check list<\/a>.<\/span><\/li>\n<li><span data-contrast=\"auto\"> If the student passes they can operate the microscope independently.<\/span><\/li>\n<li>If the student has deficiencies or is uncomfortable operating the microscope independently further training sessions with the core staff and reassessment is possible.<\/li>\n<\/ol>\n<h3><b><span data-contrast=\"auto\">Youtube Channel<\/span><\/b><\/h3>\n<p>The <a href=\"https:\/\/www.youtube.com\/@UNC-CH-CryoEM-Core\">UNC-CH CryoEM Core YouTube Channel<\/a> is committed to providing educational content related to biological cryo-electron microscopy (cryoEM), encompassing specimen preparation and the operation of the electron microscope. We post content on cryoTEM of nanoparticles, SP cryoEM of macromolecular complexes, CryoET, and microED. The content is mainly for students, post-doctoral fellows, and researchers utilizing our core facility. Our goal is to provide information to individuals interested in learning the technical aspects of cryoEM.
