Translating CF therapeutic strategies from basic research to clinical studies requires the assessment of drug candidates in physiologically relevant assays. To support translational CF research, the <b><i>CFTR Functional Analysis Core <\/i><\/b>pursues four objectives:<\/p>\n<table class=\"plain\" border=\"0\">\n<tbody>\n<tr>\n<th><img loading=\"lazy\" decoding=\"async\" class=\"alignnone wp-image-1300 size-large\" src=\"https:\/\/www.med.unc.edu\/marsicolunginstitute\/wp-content\/uploads\/sites\/547\/2015\/09\/Ussing-Chamber-Systems-1024x435.png\" alt=\"Photograph of the Ussing chamber system\" width=\"1024\" height=\"435\" srcset=\"https:\/\/www.med.unc.edu\/marsicolunginstitute\/wp-content\/uploads\/sites\/547\/2015\/09\/Ussing-Chamber-Systems-1024x435.png 1024w, https:\/\/www.med.unc.edu\/marsicolunginstitute\/wp-content\/uploads\/sites\/547\/2015\/09\/Ussing-Chamber-Systems-300x127.png 300w, https:\/\/www.med.unc.edu\/marsicolunginstitute\/wp-content\/uploads\/sites\/547\/2015\/09\/Ussing-Chamber-Systems-768x326.png 768w, https:\/\/www.med.unc.edu\/marsicolunginstitute\/wp-content\/uploads\/sites\/547\/2015\/09\/Ussing-Chamber-Systems.png 1027w\" sizes=\"auto, (max-width: 1024px) 100vw, 1024px\" \/><\/th>\n<\/tr>\n<tr>\n<td>\n<p align=\"center\">Ussing Chamber Systems for measuring electrophysiological characteristics of tissue samples and cell cultures.<\/p>\n<\/td>\n<\/tr>\n<\/tbody>\n<\/table>\n<p><b>1) <\/b>Analyze ion channel properties and correction efficiency of human bronchial epithelial (HBE) cells from harvested CF lungs to provide a full characterization report to investigators.<\/p>\n<p><b>2) <\/b>Measure ion transport function of CFTR and ENaC in HBE and human nasal epithelial (HNE) cultures by bioelectric and organoid assays to asses efficacy of candidate therapies.<\/p>\n<p><b>3)<\/b> Evaluate CFTR expression and processing by biochemical analyses to assess efficacy of CFTR modulation strategies.<\/p>\n<p><b>4)<\/b> Validate the suitability of reagents, supplies, and techniques for optimizing HBE and HNE cell integrity.<\/p>\n<p>Although numerous investigators using services provided by the <b><i>CFTR Functional Analysis Core <\/i><\/b>focus on restoration of CFTR function in CF cells by small-molecule pharmacological interventions, our services offer broad applications to multiple research programs that examine gene targeting\/therapy, modifier genes, and impact of environmental factors such as inflammation, infection, and smoking on epithelial ion transport.
