SUMMARY:MHI SEMINAR SERIES: Jonathan Brown\, M.D.\, Associate Professor\, Medicine\, Cardiovascular Medicine at Vanderbilt University
DESCRIPTION:Jonathan Brown\, MD\, is a cardiologist and basic scientist studying transcriptional mechanisms relevant to the pathogenesis of obesity\, diabetes mellitus\, and atherosclerosis. A central premise of his research involves a deeper understanding of the gene regulatory networks controlling disease pathogenesis to identify novel therapeutic targets to treat these prevalent and often fatal conditions. \nMore specifically\, Dr. Brown aims to study how chromatin regulators—as determinants of enhancer function and transcription—are involved in driving pathologic gene expression programs that alter vascular cell and adipose tissue function and result in cardiovascular disease and metabolic dysfunction. Because of his interest in the interplay among inflammation\, metabolic disease\, and atherogenesis\, he is pursuing overlapping areas of investigation examining: how proinflammatory activation of vascular endothelium and macrophages drives enhancer remodeling and gene activation through the bromodomain and extra-terminal (BET) bromodomain proteins (BRD2\, BRD3 and BRD4); and how BET proteins control adipocyte plasticity and differentiation relevant for obesity and systemic metabolic diseases. \nDr. Brown’s areas of expertise are general cardiology\, preventive cardiology\, and management of hyperlipidemia.

SUMMARY:Rong Wang\, Ph.D.\, Professor of Surgery\, Director\, Laboratory for Accelerated Vascular Research\, UCSF
DESCRIPTION:Research Overview:\n\n\nMOLECULAR PROGRAMMING OF THE VASCULATURE IN DEVELOPMENT AND DISEASE \nProper formation and function of the vasculature are crucial for health and survival\, as the vasculature supplies all cells in the body. A dysfunctional vasculature causes myriad diseases\, including stroke\, arterial occlusive diseases\, and vascular anomalies. Our long-term goal is to identify novel drug targets and inform rational therapeutic designs to treat vascular diseases. Our strategy is to understand genes crucial for angiogenesis (new vessel formation) in the normal and diseased states\, concentrating on the Notch\, ephrin-B2\, and TGF- pathways. We employ cutting-edge mouse genetics to delete or express genes in a cell lineage-specific and temporally controllable fashion in vascular cells. We combine these molecular approaches with mouse models of diseases as well as live 5D two-photon imaging (3D + blood flow over time) to uncover both the molecular mechanisms and hemodynamic signals in development and disease progression. These preclinical animal studies are coupled with patient sample validations. Our lab members come from diverse fields\, including biology\, bioengineering\, and medicine\, creating a collaborative and exciting environment. We strive to advance multiple projects across disciplines.
