In the August 5th edition of Nature Immunology, Jenny Ting’s group reports the identification of a novel NLR protein that inhibits an important inflammatory pathway. NLRs normally have been identified as activators of inflammation; however, NLRC3 binds to TLR signaling adaptor TRAF6 to form a “TRAFasome” complex which inhibits NFkB activation. This appears to be an important regulatory mechanism in endotoxic shock and sepsis. The lead author is former M&I graduate student Monika Schneider, and co-authors include current M&I graduate student Reid Roberts, former graduate student Eda Holl, and current postdocs Haitao Wen, Coy Allen, Tim Eitas, and Karen Swanson.
