{"id":2342,"date":"2023-11-08T09:38:32","date_gmt":"2023-11-08T14:38:32","guid":{"rendered":"https:\/\/www.med.unc.edu\/microscopy\/?page_id=2342"},"modified":"2024-01-30T15:25:44","modified_gmt":"2024-01-30T20:25:44","slug":"what-are-rigor-and-reproducibility","status":"publish","type":"page","link":"https:\/\/www.med.unc.edu\/microscopy\/resources\/rigor-and-reproducibility\/what-are-rigor-and-reproducibility\/","title":{"rendered":"What are rigor and reproducibility?"},"content":{"rendered":"<p><strong>Rigor<\/strong> means following procedures that will increase the likelihood of obtaining an accurate representation of the phenomenon under study.<\/p>\n<p><strong>Reproducibility <\/strong>means recording and communicating those procedures such that they can be replicated accurately.<\/p>\n<p>If your experiments are performed rigorously and reproducibly, other researchers should be able to replicate your procedures, have a high likelihood of obtaining similar results on similar samples, and have those results be an accurate representation of the phenomenon being studied.<\/p>\n<p><strong>Rigor<\/strong> requires you to pay attention to:<\/p>\n<ol>\n<li><strong>The reagents and protocols used to prepare your samples.<\/strong> MSL staff follow best practices in electron microscopy sample preparation, and can provide the exact protocols that were followed for inclusion in methods sections of papers.<\/li>\n<li><strong>The condition of the instruments used to take images and analyze images.<\/strong> MSL performs weekly maintenance on our instruments; you can review the <a href=\"https:\/\/www.med.unc.edu\/microscopy\/resources\/rigor-and-reproducibility\/msl-maintenance-procedures\/\">specific checks we perform<\/a>.<\/li>\n<li><strong>The controls included in your experiments.<\/strong> MSL provides <a href=\"https:\/\/www.med.unc.edu\/microscopy\/resources\/rigor-and-reproducibility\/light-microscopy-sample-preparation-guidelines\/\">general recommendations for controls in light-microscopy sample preparation<\/a>.<\/li>\n<li><strong>The way in which you decide which parts of your sample to study.<\/strong> This can involve decisions on which parts of samples to prepare (for example, how to section a piece of tissue, which sections to stain) and where in those samples to take images. MSL provides <a href=\"https:\/\/www.med.unc.edu\/microscopy\/resources\/rigor-and-reproducibility\/sampling\/\">suggestions for how to make sampling decisions, as well as resources for further reading<\/a>.<\/li>\n<li><strong>The way in which you decide how many images to take per sample, as well as how many samples to image.<\/strong> This mainly involves <a href=\"https:\/\/www.nature.com\/articles\/nmeth.2738\">power analysis<\/a>, a statistical technique used to determine how many samples are needed under different experimental circumstances. MSL can provide resources for researchers to do this properly.<\/li>\n<li><strong>The way in which a microscope is set up for imaging.<\/strong> MSL provides individual, detailed training with each researcher\u2019s samples to ensure they know how to properly set up imaging conditions.<\/li>\n<\/ol>\n<p><strong>Reproducibility<\/strong> requires you to pay attention to:<\/p>\n<ol>\n<li><strong>Documenting parameters used for image acquisition and saving data in proper formats.<\/strong> MSL provides advice on how to accurately record those parameters, as well as <a href=\"https:\/\/www.med.unc.edu\/microscopy\/resources\/rigor-and-reproducibility\/saving-and-opening-data-files\/\">which formats are appropriate when saving data<\/a> during our individual training sessions.<\/li>\n<li><strong>Documenting procedures used for imaging and image analysis.<\/strong> MSL can provide advice on what constitutes a proper record of those procedures.
